Lonart DS

Lonart DS (Double Strength), manufactured by Bliss GVS Pharma, is a fixed-dose Artemisinin-based Combination Therapy (ACT) antimalarial containing two synergistic active ingredients: artemether and lumefantrine.

• Artemether: A fast-acting semi-synthetic endoperoxide derivative of artemisinin. Upon interaction with intra-parasitic heme iron, it cleavage generates reactive oxygen species (free radicals) that destroy essential parasitic proteins and lipids. This produces rapid clearance of blood-stage schizonts and fast clinical symptom relief within hours.

• Lumefantrine: A long-acting lipophilic aryl-amino alcohol derivative. It acts by interfering with the polymerization process that converts toxic heme into non-toxic hemozoin inside the parasite’s food vacuole. The resulting accumulation of toxic free heme leads to parasite cell death, clearing residual parasites and preventing disease recrudescence.

The Double Strength (DS) formulation consolidates the active components into higher-strength doses (80 mg / 480 mg per tablet compared to the standard 20 mg / 120 mg), reducing the pill burden from 24 tablets down to 6 tablets total over the 3-day treatment course.

Primary Medical Indications

Lonart DS is indicated for adult and adolescent populations (weighing 35 kg or more) for the treatment of acute parasitic infections:

• Uncomplicated Plasmodium falciparum Malaria: First-line curative treatment for acute, uncomplicated malaria infections caused by P. falciparum or mixed species infections containing P. falciparum.

• Multi-Drug Resistant Malaria: Highly effective against P. falciparum strains that exhibit resistance to older antimalarials such as chloroquine, sulfadoxine-pyrimethamine (Fansidar), or quinine monotherapy.

• Treatment of Non-falciparum Blood-Stage Parasites: Effective against the erythrocytic schizonts of Plasmodium vivax, Plasmodium ovale, and Plasmodium malariae (note: radical cure of liver hypnozoites in P. vivax and P. ovale requires adjunct therapy with primaquine or tafenoquine).

• Take with Food: Always consume the tablet with a fatty meal or drink (such as milk, soup, or a meal containing oil/butter). Lumefantrine requires dietary fat for proper absorption into the bloodstream. If you vomit within 1 hour of taking a dose, take another dose immediately.

• Malaria Diagnosis: It should ideally be taken after confirmation of malaria via a Rapid Diagnostic Test (RDT) or microscopy.

• Not for Severe Malaria: Uncomplicated malaria means the patient can take oral medication and shows no signs of severe organ damage (e.g., coma, severe anemia, persistent vomiting). Severe malaria requires intravenous treatment (such as IV Artesunate) in a hospital.

• Side Effects: Common mild side effects include dizziness, fatigue, headache, loss of appetite, and joint or muscle pain.

• Cardiac Considerations: Artemether/Lumefantrine can affect cardiac rhythm (QT prolongation). Avoid taking it alongside other medications that prolong the QT interval unless advised by a doctor.

• Pregnancy: Avoid during the first trimester unless no alternative antimalarial is available and a doctor explicitly prescribes it. It is generally considered safe during the second and third trimesters under medical supervision.
